\u75c5\u6bd2\u662f\u4ec0\u9ebc\u6a23\u7684\u5b58\u5728\uff1f<\/strong><\/p>\n

\u75c5\u6bd2\uff0c\u662f\u500b\u5927\u5bb6\u8033\u719f\u80fd\u8a73\u7684\u5fae\u5c0f\u75c5\u539f\u9ad4\u3002\u6211\u5011\u66ab\u4e14\u8df3\u812b\u5c0d\u5176\u6240\u9020\u6210\u50b3\u67d3\u6027\u75be\u75c5\u7684\u6050\u61fc\u4f86\u63a2\u7a76\u75c5\u6bd2\u672c\u8cea\u4e0a\u662f\u4ec0\u9ebc\uff0c\u5b83\u57fa\u672c\u4e0a\u5c31\u662f\u4e00\u6bb5\u6709\u5305\u88dd\u7684\u907a\u50b3\u7269\u8cea\u3002\u90a3\u5305\u88dd\u53ef\u80fd\u662f\u86cb\u767d\u8cea\u6bbc\uff0c\u53ef\u80fd\u6709\u8102\u8cea\u5916\u819c\uff0c\u6216\u6bbc\u819c\u517c\u5099\uff0c\u7e3d\u4e4b\u5305\u88f9\u6210\u4e00\u500b\u5948\u7c73\u9846\u7c92\u3002\u5916\u6bbc\u6216\u819c\u4e0d\u4f46\u4fdd\u8b77\u5167\u90e8\u7684\u907a\u50b3\u7269\u8cea\uff0c\u4e5f\u5354\u52a9\u75c5\u6bd2\u907a\u50b3\u7269\u8cea\u9032\u5165\u5bbf\u4e3b\u7d30\u80de\u5167\u3002\u800c\u5176\u5167\u542b\u7684\u907a\u50b3\u7269\u8cea\u53ef\u80fd\u662f DNA\u3001\u6216\u662f\u540c\u6a23\u53ef\u50b3\u905e\u907a\u50b3\u8a0a\u606f\u7684 RNA\uff0c\u7e3d\u4e4b\u75c5\u6bd2\u7684\u4efb\u52d9\u5c31\u662f\u628a\u81ea\u5df1\u7684 DNA \u6216 RNA \u9001\u5230\u5bbf\u4e3b\u7d30\u80de\u4e2d\uff0c\u81f4\u4f7f\u5bbf\u4e3b\u7d30\u80de\u4f9d\u7167\u75c5\u6bd2\u9001\u4f86\u7684\u6307\u793a\u751f\u7522\u66f4\u591a\u65b0\u751f\u7684\u75c5\u6bd2\u3002<\/p>\n

\u75c5\u6bd2\u4e0d\u4e00\u5b9a\u8981\u8b93\u5bbf\u4e3b\uff08\u6211\u5011\uff09\u751f\u75c5\uff0c\u4f46\u82e5\u5728\u8907\u88fd\u75c5\u6bd2\u904e\u7a0b\u4e2d\u3001\u6216\u5bbf\u4e3b\u6297\u62d2\u6642\u9020\u6210\u5bbf\u4e3b\u751f\u75c5\u4e86\uff0c\u53ea\u8981\u4e0d\u5f71\u97ff\u75c5\u6bd2\u8907\u88fd\uff0c\u5c0d\u75c5\u6bd2\u4f86\u8aaa\u7686\u7121\u59a8\u3002\u75c5\u6bd2\u9846\u7c92\u88e1\u5305\u88f9\u7684\u907a\u50b3\u7269\u8cea\u82e5\u662f DNA\uff0c\u90a3\u9ebd\u8907\u88fd\u904e\u7a0b\u4e2d\u75c5\u6bd2 DNA \u6703\u5148\u88ab\u8f49\u9304\u6210 RNA \u518d\u8f49\u8b6f\u70ba\u75c5\u6bd2\u8907\u88fd\u6216\u7d44\u88dd\u6240\u9700\u7684\u86cb\u767d\u8cea\u3002\u53e6\u6709\u5f88\u591a\u75c5\u6bd2\u662f\u76f4\u63a5\u5305\u88f9 RNA \u7684\uff0c\u90a3\u9ebc\u75c5\u6bd2 RNA \u4e5f\u53ef\u4ee5\u88ab\u8f49\u8b6f\u70ba\u86cb\u767d\u8cea\u3002\u7e3d\u4e4b\u75c5\u6bd2\u5728\u8907\u88fd\u904e\u7a0b\u4e2d\u4e00\u5b9a\u6709\u500b\u6b65\u9a5f\u6703\u9700\u8981\u7528\u5230 RNA\u3002\u56e0\u6b64\uff0c RNA \u4e0d\u53ea\u662f\u5206\u5b50\u751f\u7269\u5b78\u7684\u8cc7\u8a0a\u50b3\u905e\u93c8\u4e2d\uff08DNA \u2192 RNA \u2192\u86cb\u767d\u8cea\uff09\u7684\u4e2d\u5805\u4efd\u5b50\uff0c\u4e5f\u662f\u75c5\u6bd2\u8907\u88fd\u904e\u7a0b\u4e2d\u6240\u5fc5\u9808\u4f7f\u7528\u5230\u7684\u5206\u5b50\u7a2e\u985e\u3002<\/p>\n

\u907a\u50b3\u7269\u8cea RNA \u5e8f\u5217\u4e0a\u7684\u984d\u5916\u5316\u5b78\u4fee\u98fe<\/strong><\/p>\n

\u57fa\u790e\u751f\u7269\u5b78\u4e2d\u666e\u904d\u5c0d RNA \u7684\u4ecb\u7d39\u662f\uff1a\u751f\u7269\u7684\u57fa\u56e0\u8a18\u9304\u65bc DNA \u4e4b\u4e2d\uff0c\u800c\u9019\u4e9b\u57fa\u56e0\u8981\u8868\u73fe\u3001\u4f7f\u5176\u6709\u529f\u80fd\uff0c\u6703\u9700\u8981\u5148\u5f9e DNA \u628a\u8a0a\u606f\u6284\u5beb\uff08\u8f49\u9304\uff09\u70ba RNA\uff0cRNA \u4e0a\u7684\u8a0a\u606f\u518d\u88ab\u8f49\u8b6f\u70ba\u6709\u529f\u80fd\u7684\u86cb\u767d\u8cea\u3002\u5982\u679c\u6211\u5011\u628a\u9019\u904e\u7a0b\u6bd4\u55bb\u70ba\u4e00\u500b\u5eda\u5e2b\u5728\u505a\u83dc\uff0c\u90a3\u5eda\u5e2b\u5148\u770b\u7684\u98df\u8b5c\u662f DNA\uff0c\u7528\u4f86\u6284\u5beb\u7b46\u8a18\u7684\u4fbf\u689d\u7d19\u5373\u70ba RNA\uff0c\u800c\u4f9d\u7167\u4fbf\u689d\u7d19\u505a\u51fa\u4f86\u7684\u6599\u7406\u5c31\u662f\u300c\u6709\u529f\u80fd\u7684\u86cb\u767d\u8cea\u300d\u3002<\/p>\n

\u9577\u4e45\u4ee5\u4f86\uff0c\u751f\u7269\u5b78\u754c\u4e00\u76f4\u8a8d\u70ba\u627f\u8f09\u8457\u57fa\u56e0\u5e8f\u5217\u7684\u907a\u50b3\u7269\u8cea\uff0c\u4e0d\u7ba1\u662f A\u3001T\u3001C\u3001G \u56db\u500b\u9e7c\u57fa\u7d44\u5408\u800c\u6210\u7684 DNA\uff0c\u9084\u662f A\u3001U\u3001C\u3001G \u7d44\u6210\u7684 RNA \u90fd\u662f\u53ea\u7528\u56db\u500b\u9e7c\u57fa\u7684\u6392\u5217\u7d44\u5408\u5c31\u53ef\u4ee5\u8a18\u9304\u3001\u50b3\u905e\u751f\u547d\u5ef6\u7e8c\u8207\u904b\u4f5c\u6240\u9700\u7684\u8cc7\u8a0a\u4e86\u3002\u4e0d\u904e\u8fd1\u5e7e\u5e74\u4f86\uff0c\u4eba\u5011\u9010\u6f38\u77ad\u89e3\u5230 DNA \u4e26\u975e\u5168\u90e8\u4e00\u5b9a\u6703\u88ab\u8b80\u5230\uff0c\u800c\u662f\u4e0d\u540c\u5340\u6bb5\u6703\u88ab\u984d\u5916\u6a19\u8a18\u6b64\u6bb5\u662f\u5426\u9700\u8981\u88ab\u89e3\u8b80\u8207\u4f7f\u7528\u3002\u9019\u4e9b\u984d\u5916\u7684\u6a19\u8a18\uff0c\u4f8b\u5982\uff1a\u5728 DNA \u4e0a\u52a0\u4e0a\u7532\u57fa\uff08CH3<\/sub>, methylation\uff09\u4fee\u98fe\uff0c\u5c0d DNA \u88ab\u89e3\u8b80\u8207\u5426\u7684\u8abf\u63a7\uff0c\u5373\u6210\u70ba\u4e86\u300c\u8868\u89c0\u907a\u50b3\u5b78\uff08epigenetics\uff09\u300d\u9019\u500b\u65b0\u8208\u5b78\u9580\u3002\u540c\u6a23\u7684\uff0cRNA \u4e5f\u53ef\u4ee5\u88ab\u4fee\u98fe\uff0c\u9032\u800c\u8abf\u63a7 RNA \u4e4b\u529f\u80fd\u3001\u8f49\u8b6f\u3001\u8207\u5b58\u5728\u6642\u9593\uff0c\u6b64\u7a31\u70ba\u300c\u8868\u89c0\u8f49\u9304\u9ad4\u5b78\uff08epitranscriptomics\uff09\u300d\u3002<\/p>\n

\u65e9\u671f\u767c\u73fe\u7684\u75c5\u6bd2 RNA \u4fee\u98fe<\/strong><\/p>\n

\u56e0\u70ba\u75c5\u6bd2\u5c0f\uff0c\u5b83\u5011\u7684\u57fa\u56e0\u9ad4\u5e8f\u5217\u6bd4\u5927\u90e8\u5206\u751f\u7269\u7684\u77ed\u5f88\u591a\uff0c\u56e0\u6b64\u5728\u8655\u7406\u9577\u5e8f\u5217\u7684\u6280\u8853\u76f8\u5c0d\u5c11\uff0c\u7814\u7a76\u65e9\u671f\uff0c\u75c5\u6bd2\u6210\u70ba\u5f88\u591a\u5206\u5b50\u751f\u7269\u5b78\u300c\u76f8\u5c0d\u7c21\u55ae\u300d\u7684\u7814\u7a76\u4e3b\u984c\u3002\u4ee5 RNA \u4fee\u98fe\u4f86\u8aaa\uff0c\u65e9\u5728 1975 \u5e74 Lavi & Shatkin \u5373\u767c\u73fe\u733f\u7334\u75c5\u6bd2 40\uff08SV40\uff09\u7684 RNA \u5728 A \u7684\u9e7c\u57fa\u4e0a\uff08\u817a\u82f7\uff09\u6709\u7532\u57fa\u5316\u4fee\u98fe\uff08N6-methyladenosine, m6A\uff09\u3002\u7dca\u63a5\u8457\u5728 1976 \u5230 1977 \u5e74\u9593 A \u578b\u6d41\u611f\u75c5\u6bd2\uff08Influenza A virus\uff09\u3001\u817a\u75c5\u6bd2\uff08Adenovirus\uff09\u3001\u52de\u65af\u8089\u7624\u75c5\u6bd2\uff08Rous sarcoma virus\uff09\u7684 RNA \u7686\u88ab\u767c\u73fe\u6709\u7532\u57fa\u5316\u4fee\u98fe\u7684\u73fe\u8c61\u3002\u4e0d\u904e\u7919\u65bc\u6280\u8853\u7684\u9650\u5236\uff0c\u75c5\u6bd2\u5b78\u754c\u9577\u671f\u7121\u6cd5\u7834\u89e3\u75c5\u6bd2 RNA \u4fee\u98fe\u7684\u529f\u80fd\u70ba\u4f55\u3002<\/p>\n

\u75c5\u6bd2 RNA \u4fee\u98fe\u53ef\u589e\u5f37\u75c5\u6bd2\u7684\u57fa\u56e0\u8868\u73fe<\/strong><\/p>\n

\u76f4\u5230\u8fd1\u671f\u65b0\u6280\u8853\u7684\u958b\u767c\u8207 RNA \u4fee\u98fe\u9175\u7d20\u7684\u767c\u73fe\uff0c\u81f4\u4f7f\u75c5\u6bd2 RNA \u4fee\u98fe\u7684\u7814\u7a76\u518d\u5ea6\u6d3b\u7d61\u4e86\u8d77\u4f86\uff0c\u9010\u6b65\u63ed\u958b\u75c5\u6bd2 RNA \u4fee\u98fe\u7684\u795e\u79d8\u9762\u7d17\u3002\u5f9e 2016 \u5e74\uff0c\u4e09\u500b\u5be6\u9a57\u5ba4\u540c\u6642\u767c\u73fe\u4eba\u985e\u514d\u75ab\u7f3a\u4e4f\u75c5\u6bd2\uff08HIV-1\uff09\u7684 RNA \u4e5f\u6709 m6<\/sup>A \u7532\u57fa\u5316\u4fee\u98fe\u958b\u59cb\uff0cRNA \u4fee\u98fe\u9010\u6f38\u88ab\u767c\u73fe\u4e26\u7cbe\u6e96\u5b9a\u4f4d\u65bc\u591a\u7a2e\u75c5\u6bd2\u7684 RNA \u4e0a\uff0c\u4f8b\u5982\uff1a\u65e5\u672c\u8166\u708e\u75c5\u6bd2\u3001B \u578b\u809d\u708e\u75c5\u6bd2\u3001C \u578b\u809d\u708e\u75c5\u6bd2\u3001\u547c\u5438\u9053\u878d\u5408\u75c5\u6bd2\uff08Respiratory syncytial virus, RSV\uff09\u3001\u4ee5\u53ca\u5404\u7a2e\u51a0\u72c0\u75c5\u6bd2\u7b49\uff0c\u7686\u76f8\u7e7c\u88ab\u767c\u73fe\u5176 RNA \u4e0a\u6709\u7532\u57fa\u5316\u4fee\u98fe\u3002\u6709\u8da3\u7684\u662f\uff0c\u5927\u90e8\u5206\u75c5\u6bd2\u7684 RNA \u90fd\u662f\u9700\u8981\u88ab\u4fee\u98fe\u7684\uff0c\u53ea\u8981\u53bb\u9664\u4e86 RNA \u4fee\u98fe\uff0c\u5f88\u591a\u75c5\u6bd2\u7684\u8907\u88fd\u6548\u7387\u7686\u6703\u986f\u8457\u4e0b\u964d\u3002<\/p>\n

RNA \u7532\u57fa\u5316\u4fee\u98fe\u4fc3\u9032\u75c5\u6bd2\u8907\u88fd\u7684\u73fe\u8c61\uff0c\u5728 HIV-1 \u548c\u817a\u75c5\u6bd2\u4e2d\u5c24\u5176\u660e\u78ba\uff0c\u6709\u4fee\u98fe\u7684 HIV-1 RNA \u53ef\u4ee5\u5728\u5bbf\u4e3b\u7d30\u80de\u4e2d\u5b58\u6d3b\u6bd4\u8f03\u4e45\uff0c\u907f\u514d\u75c5\u6bd2 RNA \u5728\u5b8c\u6210\u4efb\u52d9\u524d\u5148\u88ab\u5bbf\u4e3b\u7d30\u80de\u88c2\u89e3\uff1b\u6b64\u5916 RNA \u53ef\u4ee5\u5728\u751f\u7522\u51fa\u4f86\u5f8c\u518d\u88ab\u526a\u63a5\uff08RNA splicing\uff09\uff0c\u4e0d\u540c\u7684\u526a\u63a5\u65b9\u5f0f\u8868\u73fe\u51fa\u4e0d\u540c\u7684\u86cb\u767d\u8cea\uff0c\u800c RNA \u7532\u57fa\u5316\u53ef\u4ee5\u8abf\u63a7 RNA \u7684\u526a\u63a5\uff0c\u4ee5\u78ba\u4fdd HIV-1 \u548c\u817a\u75c5\u6bd2\u8868\u73fe\u7684\u75c5\u6bd2\u86cb\u767d\u8cea\u7a2e\u985e\u6b63\u78ba\u3001\u6216\u5404\u7a2e\u985e\u8868\u73fe\u6bd4\u4f8b\u5408\u4e4e\u75c5\u6bd2\u7576\u4e0b\u7684\u9700\u6c42\u3002<\/p>\n

RNA \u4fee\u98fe\u53ef\u8abf\u63a7\u7d30\u80de\u7684\u6297\u75c5\u6bd2\u514d\u75ab<\/strong><\/p>\n

\u9664\u4e86\u8abf\u63a7\u75c5\u6bd2\u57fa\u56e0\u7684\u8868\u73fe\uff0cRNA \u4fee\u98fe\u4e5f\u53ef\u4ee5\u8abf\u63a7\u5bbf\u4e3b\u7d30\u80de\u7684\u6297\u75c5\u6bd2\u53cd\u61c9\u3002\u4eba\u985e\u5de8\u7d30\u80de\u75c5\u6bd2\uff08Human Cytomegalovirus, HCMV\uff09\u662f\u500b\u4e0d\u5e38\u807d\u5230\uff0c\u4f46\u53ef\u9020\u6210\u65b0\u751f\u5152\u807d\u529b\u56b4\u91cd\u53d7\u640d\u7684\u5e38\u898b\u75c5\u6bd2\u3002\u96d6\u7136\u4eba\u985e\u5de8\u7d30\u80de\u75c5\u6bd2\u7684 RNA \u4e0a\u627e\u4e0d\u5230\u7532\u57fa\u5316\u4fee\u98fe\uff0c\u4f46\u9019\u75c5\u6bd2\u53ef\u4ee5\u589e\u5f37\u5bbf\u4e3b\u7d30\u80de\u5167\u7684 RNA \u7532\u57fa\u5316\u6a5f\u5236\uff0c\u4f7f\u7d30\u80de\u5167\u7684\u91cd\u8981\u6297\u75c5\u6bd2\u57fa\u56e0\u5e72\u64fe\u7d20 \u03b2\uff08interferon \u03b2\uff09\u7684 RNA \u88ab\u7532\u57fa\u5316\u3002\u6709\u8da3\u7684\u662f\u5bbf\u4e3b\u7d30\u80de\u7684 RNA \u88ab\u7532\u57fa\u5316\u53cd\u800c\u53ef\u4ee5\u4fc3\u9032\u5176\u88c2\u89e3\uff0c\u81f4\u4f7f\u9019\u75c5\u6bd2\u611f\u67d3\u904e\u7a0b\u4e2d\uff0c\u501a\u8cf4\u5e72\u64fe\u7d20 \u03b2 \u7684\u6297\u75c5\u6bd2\u6a5f\u5236\u88ab\u524a\u5f31\uff0c\u4f7f\u75c5\u6bd2\u80fd\u5920\u66f4\u6709\u6548\u7387\u7684\u8907\u88fd\u3002<\/p>\n

2005 \u5e74 Kariko & Weissman \u767c\u73fe\u4eba\u5de5\u5408\u6210\u7684 RNA \u82e5\u76f4\u63a5\u9001\u9032\u7d30\u80de\u88e1\uff0c\u6703\u8a98\u767c\u7522\u751f\u56b4\u91cd\u7684\u514d\u75ab\u53cd\u61c9\u800c\u9020\u6210\u7d30\u80de\u6b7b\u4ea1\uff0c\u82e5\u52a0\u5165\u7532\u57fa\u5316\u4fee\u98fe\u5247\u53ef\u4ee5\u907f\u514d\u4eba\u5de5 RNA \u88ab\u7d30\u80de\u5075\u6e2c\u70ba\u5916\u4f86\u7269\u7522\u751f\u514d\u75ab\u53cd\u61c9\uff0c\u5728\u5404\u7a2e\u4fee\u98fe\u4e2d\u4ee5\u7532\u57fa\u5316\u5047\u5c3f\u5627\u5576\u6838\u82f7\uff08N1<\/sup>-methylpseudouridine, m1\u03c8\uff09\u7684\u6548\u679c\u6700\u597d\u3002\u6b64\u767c\u73fe\u4f7f\u4eba\u5011\u7406\u89e3\u5230\u52a0\u5165\u7532\u57fa\u5316\u4fee\u98fe\u5373\u53ef\u53bb\u9664\u4eba\u5de5\u5408\u6210 RNA \u5c0d\u7d30\u80de\u7684\u6bd2\u6027\uff0c\u800c\u61c9\u7528\u6b64\u6280\u8853\u7684 RNA \u75ab\u82d7\u6210\u70ba\u4e86\u5168\u4e16\u754c COVID-19 \u80ba\u708e\u75ab\u60c5\u7684\u89e3\u65b9\uff0c\u56e0\u6b64\u4eca\u5e74\uff082023\uff09Kariko & Weissman \u7372\u9812\u4e86\u8afe\u8c9d\u723e\u91ab\u5b78\u734e\u3002<\/p>\n
